Reagent for measuring magnesium ion

Abstract

PURPOSE: The titled reagent, consisting of glucokinase, adenosine triphosphate, glucose, glucose-6-phosphate dehydrogenase and oxidized form nicotinamide adenine dinucleotide phosphate and having simple operation and high specificity and accuracy. CONSTITUTION: A reagent obtained by preparing the first reagent consisting of 1mM adenosine triphosphate (ATP), 0.5mM oxidized form nicotinamide adenine dinucleotide phosphate (NADP), 30mM glucose and 50mM tris-hydrochloric acid buffer solution (8.5pH) and the second reagent consisting of 0.65U/ml glucokinase derived from Bacillus stearothermophilus and 0.65U/ml glucose-6- phosphate dehydrogenase derived from Leuconostock mesenteroides, mixing 2.4ml of the first reagent with 0.01ml Mg standard solution, preheating the mixture solution at 30°C for 3min, adding 0.6ml of the second reagent, measuring the change in absorbance at 340nm for 3min and obtaining a calibration curve for measuring Mg.
